實(shí)驗(yàn)原理:將一種抗mulv p30單克隆包衣抗體吸附在微量滴定板上。樣品或標(biāo)準(zhǔn)品中存在的mulv核心抗原(p30)與吸附在板上的抗體結(jié)合;添加抗mulv p30多克隆抗體并與抗體捕獲的抗原結(jié)合。在孵育和洗滌步驟后,添加一種HRP結(jié)合的二級(jí)抗體并結(jié)合到抗mulv p30多克隆。在洗滌過(guò)程中去除未結(jié)合的HRP結(jié)合的二級(jí)抗體,并將與HRP反應(yīng)的底物溶液加入到空中。有色產(chǎn)物的形成與樣品中存在的mulv核心抗原的量成比例。通過(guò)添加酸終止反應(yīng),并在450 nm處測(cè)量吸光度。用重組mulv p30核心抗原制備標(biāo)準(zhǔn)曲線,測(cè)定樣品濃度。
